Research Summary:One of the limitations of studying enzymatic systems is the availability of a simple spectrophotometric assay that can be used to determine kinetic parameters and inhibition constants by monitoring the absorbance or fluorescence of the chromophoric products of the enzymatic reactions. Furthermore, these methods are known to be labor-intensive and time-consuming.

The Leary Lab has developed a fast and accurate enzyme assay that allows kinetic parameters determination and inhibition studies of nonchromophoric systems using Electrospray Ionization Mass Spectrometry (ESI-MS). My project focuses on using this technique, further develop and improve it, on enzyme systems that have biological significances to calculate their respective Km, Vmax, and Kis, and determine the mechanisms of their reactions..
